Live & DeadTM動(dòng)物細(xì)胞活力/毒性檢測(cè)試劑盒(Calcein AM, EthD-I)是一種為動(dòng)物細(xì)胞死活檢測(cè)提供雙熒光染色的試劑盒。試劑盒內(nèi)的兩種探針可分別測(cè)定細(xì)胞內(nèi)酯酶活性和質(zhì)膜完整性從而反映細(xì)胞活力。本試劑盒可用于熒光顯微鏡、流式細(xì)胞儀、酶標(biāo)儀以及其他熒光檢測(cè)系統(tǒng)。
本試劑盒可以應(yīng)用于大多數(shù)的真核哺乳動(dòng)物細(xì)胞,包括貼壁細(xì)胞核的某些組織,但不適用于真菌和酵母。該試劑盒與相同用處的臺(tái)盼藍(lán)相比,更快捷安全且靈敏度更高。
注意事項(xiàng)
1. 使用前請(qǐng)將產(chǎn)品瞬時(shí)離心至管底,再進(jìn)行后續(xù)實(shí)驗(yàn)。
2. 酚紅或血清對(duì)于本試劑盒的檢測(cè)有一定的干擾。
3. 熒光染料均存在淬滅問(wèn)題,實(shí)驗(yàn)操作時(shí)請(qǐng)盡量注意避光,以減緩熒光淬滅。

常見(jiàn)問(wèn)題解答:
◆你們家有兩款哺乳動(dòng)物死活染色試劑盒,分別是Calcein AM,PI(貨號(hào)L6037)與Calcein AM,EthD-1(貨號(hào)L6023,這兩者的區(qū)別是什么?
EthD-1相較于PI來(lái)說(shuō),對(duì)DNA的親和力更高,低濃度下即可滿(mǎn)足染色效果。
◆我如何為L(zhǎng)IVE/DEAD細(xì)胞活力實(shí)驗(yàn)準(zhǔn)備死細(xì)胞對(duì)照?
有兩種簡(jiǎn)單的方法。一種是通過(guò)60°C放置20分鐘熱滅活細(xì)胞。第二種是將細(xì)胞放入70%乙醇。乙醇固定的細(xì)胞可以在冰箱中無(wú)限期儲(chǔ)存直到使用,可達(dá)好幾年。
步驟:
a.離心細(xì)胞、使細(xì)胞沉淀并去除上清
b.固定細(xì)胞:在15mL含有細(xì)胞團(tuán)的試管中加入10 mL70%冰乙醇,先開(kāi)始逐滴加入后震蕩,混合均勻。
c.在冰箱中儲(chǔ)存直到使用。
d.快使用時(shí),水洗兩次且在選擇的緩沖液中重懸。
◆用該試劑盒染色后,可以進(jìn)行后續(xù)固定步驟嗎?
染色后不可以進(jìn)行固定步驟。PI作為膜非透性染料,固定后將轉(zhuǎn)移到所有細(xì)胞,不再維持死細(xì)胞特異性染色。
